欧李休眠相关基因MADS-box5 (DAM5)的功能分析

摘 要:

Dormancy-associated MADS-box (DAM)基因在植物休眠中有着重要作用, 欧李中尚未对该类基因进行功能研究。本文通过构建ChDAM5过表达载体、实时荧光定量PCR以及拟南芥遗传转化等技术, 分析相关基因的表达及脱落酸含量变化。结果表明, ChDAM5基因在拟南芥中的过表达激活了拟南芥脱落酸合成相关基因(AtNCED6AtNCED9)的表达, 促进脱落酸合成, 最终抑制了种子萌发.

Functional analysis of DAM5 gene in Cerasus humilis

Abstract:

In the study of dormancy genes, dormancy associated MADS box (DAM) gene plays an important role in dormancy, but the function of this kind of gene has not been studied in Cerasus humilis. In this paper, the expression of related genes and the changes of abscisic acid content were analyzed by constructing the overexpression vector of ChDAM5, real-time fluorescent quantitative PCR and genetic transformation of Arabidopsis thaliana. The results showed that the overexpression of ChDAM5 gene in Arabidopsis  activated the expression of Arabidopsis ABA synthesis related genes (AtNCED6 and AtNCED9), promoted ABA synthesis, and fnally inhibited seed germination.
